It is well known that oxidative stress is directly responsible for a variety of diseases and disorders, including diabetes, hypertension, and atherosclerosis. Hydrogen has been found to have a strong oxidative stress reducing effect. Therefore, we investigated the effects of water intake enriched with molecular hydrogen (H2) on lipid metabolism and glucose levels in patients with type 2 diabetes mellitus (T2DM) or impaired glucose tolerance (IGT). We performed a randomized, double-blind, placebo-controlled, crossover study in 30 patients with type 2 diabetes and 6 patients with impaired glucose tolerance controlled by diet and exercise therapy. Patients ingested either 900 mL/day of hydrogen water or 900 mL/day of pure placebo water for 8 weeks, with a 12-week washout period. Biomarkers of oxidative stress, insulin resistance, and glucose metabolism were examined by oral glucose tolerance test at baseline and at week 8. Hydrogen water supplementation resulted in significant decreases in modified low-density lipoprotein (LDL) cholesterol levels (ie, modifications that increase the net negative charge of LDL), small dense LDL, and urinary 8-isoprostanes by 15.5% (P <.01), 5.7% (P <.05), and 6.6% (P <.05) in patients, respectively. Intake of H2-rich water also resulted in a trend toward lower serum concentrations of oxidized LDL and free fatty acids and increased plasma levels of adiponectin and extracellular superoxide dismutase. In 4 of 6 patients with impaired glucose tolerance, intake of hydrogen water normalized glucose levels at baseline tolerance test. In conclusion, these results establish that hydrogen water intake improves the condition of patients with type 2 diabetes, as well as having beneficial effects in preventing type 2 diabetes and insulin resistance.
